Sorting nexin-5 is integral to various stages of intracellular trafficking, notably through its interaction with membranes rich in phosphatidylinositol 3-phosphate or phosphatidylinositol 3,4-bisphosphate. As part of the retromer SNX-BAR subcomplex, it is essential for the retrograde transport of proteins to the trans-Golgi network and plays a role in recycling cargo proteins from the plasma membrane. It is involved in the transport of IGF2R, the internalization of EGFR following EGF stimulation, and the sorting and degradation of EGFR and E-cadherin, highlighting its significance in cellular signaling and protein degradation pathways.
